The most interesting part of the monastery is the "defense church" of the Annunciation of the Blessed Virgin Mary. Originally built between 1503 and 1511, it was one of the few churches of its kind on the Byzantine periphery—architecturally combining elements of Western Gothic and Eastern Byzantine culture. On July 21, 1944, the church, along with most of the significant buildings in Supraśl, was destroyed by German troops. Reconstruction of the church, based on the preserved foundations, took over 30 years, and it was finally consecrated in 2021.

The five-domed Church of St. Peter and Paul, built in 1853 in the Byzantine-Russian style, features a magnificent gateway serving as a bell tower, added in 1873. A feature of the church is its historic iconostasis dating from 1836, dating twenty years earlier than the church itself.
